Mechanism of Action
Anastrozole is a potent and highly selective non-steroidal aromatase inhibitor.
In postmenopausal women, estradiol is produced mainly through the conversion of androstenedione to estrone by the aromatase enzyme complex in peripheral tissues. Estrone is subsequently converted to estradiol.
Reducing circulating estradiol levels has been shown to provide therapeutic benefits in women with breast cancer. Studies have demonstrated that a daily dose of 1 mg anastrozole reduces estradiol levels in postmenopausal women by more than 80%.
Anastrozole has no progestogenic, androgenic, or estrogenic activity.
Clinical studies have also shown that doses up to 10 mg daily do not affect cortisol or aldosterone secretion. Therefore, corticosteroid replacement therapy is not required.
